Upgrading - Will I lose all my customizations?
Greetings. I'm using an older version of WordPress and the Hypershot theme. If I upgrade to 4.2.1 and install the newest update of the theme, will I lose my customizations? Just want to figure out how much work is ahead of me.

If you mean settings in theme options, then you wont lose them.
If you made changes in theme templates, and you didn't do them in child theme then yes, you will lose such changes as theme is overwritten fully.
If you made same changes but in child theme, then most of time you should be safe. Only when there was some new feature added in place that you have changed.
